TRYPTYR (acoltremon ophthalmic solution) 0.003% is a prescription eye drop used for the treatment of the signs and symptoms of dry eye disease (DED).

Narrator: To get ready, wash your hands and remove contacts if you're wearing them. Simply pull one single-dose vial from the strip.

Narrator: Now tilt your head back, gently pull your lower eyelid downwards and look up. Some people may find having a mirror in front of them to be helpful.

Narrator: Place the tip of the vial close to your eye, but do not let it touch your eye.
Narrator: Gently squeeze the vial and let one full drop of TRYPTYR coat your eye, then gently close your eye for a few seconds. You then use the same vial to apply one drop to your other eye, the same way, keeping your head tilted back and your eyelid held down.
You may feel a sensation. In clinical trials, half of patients using TRYPTYR reported burning or stinging. 1.2% reported it as moderate or severe. In another safety study, where they measured the duration, 86% of patients experiencing burning or stinging reported it lasted a minute or less.

Use TRYPTYR exactly as your healthcare provider tells you to.
TRYPTYR is for use in the eye. Each single-dose vial of TRYPTYR will give you enough medicine to treat both of your eyes. TRYPTYR vial is for 1-time use only. Use right away after opening and do not reuse.
Keep the unopened single-dose vials in the original foil package until ready to use.
Wash your hands before each use. Do not let the tip of the single-dose vial touch your eye or any other surface to avoid eye injury or infection.
If you use TRYPTYR with other eye medicines, wait at least 5 minutes between using TRYPTYR and the other medicines.
If you wear contact lenses, remove them before using TRYPTYR. Wait 15 minutes after dosing before putting contact lenses back in your eyes.
Call your healthcare provider right away if you get an allergic reaction or other eye problems such as eye injury, eye infection, or eye pain.
The most common side effect of TRYPTYR in clinical trials was burning or stinging sensation in the eye upon instillation, which was reported in 50% of patients.

TRYPTYR can be a simple part of your daily routine
- Apply 1 drop to each eye 2 times a day, approximately 12 hours apart
- If using with other eye drops, apply at least 5 minutes apart
- Wait to insert contact lenses 15 minutes following application
- Call your doctor right away if you experience an allergic reaction or other eye problems such as eye injury, eye infection, or eye pain
If you missed a dose, resume regular dosing at the next scheduled time. If you misplace or run out of TRYPTYR before your next prescription, talk to your doctor. Don’t forget to review the full Instructions for Use before getting started.
Storing TRYPTYR
- Unopened cartons of TRYPTYR should be stored in a refrigerator at 2°C to 8°C (36°F to 46°F)
- After opening the carton, TRYPTYR may be stored in a refrigerator or at room temperature at 2°C to 25°C (36°F to 77°F)
- If stored at room temperature, TRYPTYR should be used within 30 days, not to exceed the expiration date printed on the carton and foil pouch
- After opening each foil pouch, the single-dose vials should be used within 7 days, not to exceed the expiration date printed on the vial. Store unopened single-dose vials in the original foil pouch until ready to use

TRYPTYR is the ONLY prescription eye drop that uses your eyes' natural processes to quickly make more of your own tears.
In dry eye, your eyes don’t make enough natural tears. TRYPTYR stimulates your nerve and tear glands that produce the components of natural tears.
TRYPTYR works fast. Natural tear production increased as early as Day 1 and was sustained over time. Patients experienced symptom relief by Day 28 in one clinical trial. A significant reduction was not observed at day 28 in the second clinical trial.*

If you miss a dose of TRYPTYR you can continue using it at the next scheduled time. For example, if you miss your morning dose, simply take TRYPTYR in the evening.

In clinical trials that included 766 patients, the most common side effect of TRYPTYR was a burning or stinging sensation after applying the eye drop. This was reported in 50% of the patients using TRYPTYR. Only 1.2% of patients reported the sensation as moderate or severe.
